Oligonucleotide therapeutics is one of the most promising ﬁelds in modern medicine to treat several diseases of genetic and non-genetic nature, by modulating gene expression. Though advancement and growth in oligo therapeutics appear stable and promising, oligos face immense challenges for their use as therapeutics and these must be resolved before proceeding with regulatory approvals. Highly complex nature of oligo drugs requires novel and sophisticated process methods including new types of equipment that still satisfy the cGMP requirements. Successful application of these agents requires appropriate design and delivery strategies to increase their stability and intracellular uptake. Relatively inordinate cost of oligos is a major concern, arising from the less coupling efficiency and the excess use of amidites and thereby the high cost incurred in oligo process development. In this session, I will present the major challenges in oligo therapeutics, which are associated with technology, manufacturing, delivery and process of regulation.
