The KxS process refractometer with 1” Ingold process connection provides real-time refractive index (RI) and concentration measurements in pilot and high-volume bioreactors, fermenters, and separation systems – facilitating mass production.
Refractive index measures the change in light speed as it passes through a material, specifically the deviation caused by differences in optical densities. This deviation occurs because it is harder for light to travel through a solution if there is more dissolved material.
In bioprocessing, RI measurements are commonly used to monitor e.g., proteins or protein buffer solutions. As dissolved solids like proteins increase, light travels more slowly, making RI directly proportional to total dissolved solids content.
RI acts as a universal detector which can follow chemical reactions or mass transfer, whilst also clearly detecting the steady state of a process step. As a Process Analytical Technology (PAT) tool, RI minimizes the need for manual sampling, which is time-consuming and labor-intensive, requiring dedicated personnel. Sampling introduces a higher risk of contamination during downstream processing.
Inline monitoring allows for continuous tracking of concentration levels, helping to prevent variations that could lead to batch inconsistencies. Operators can observe the data as it is instantly detected by the refractometer, as RI reflects what is happening in the solution. This consistency is vital for meeting regulatory standards and ensuring that the final product is safe and effective for therapeutic use.
